TT Lock - Ultrasensitive diagnostic tool

A James Cook University-based team has generated a specific DNA sequence, called TT-Lock, which allows immunoPCR amplification without the problems of non-specific binding and variable stoichiometry. The TT-Lock's partially forked 21 bp DNA sequence binds to Tus with excellent stability.

This protein-DNA interaction is the strongest of its kind reported for a monomeric DNA-binding protein with a dissociation half life of 89 min in 250 mM KCl at room temperature. The TT-Lock technology offers a fast and reliable method for the ultra-sensitive detection of low copy protein numbers, with the potential to significantly increase signal amplification in immunoPCR systems, as well as in a variety of other applications that require DNA or protein molecules to be immobilised on surfaces.
